I have run a LiveUSB with persistence on a machine continuously for months with numerous suspend-and-wake cycles without any problem. Oftentimes I removed the usb from its slot and put it back in before I woke up the machine. As a habit I always created a swap partition on the usb, though I don't think it should have made any difference.
